چکیده

The potential of line-scan Raman hyperspectral imaging system equipped with a 785 nm line laser was examined for discrimination uninfected control, AF36-inoculated and AF13-inoculated corn kernels in this study. AF13 AF36 strains were used as representatives the aflatoxigenic non-aflatoxigenic A. flavus fungi. A total 300 3 treatments, namely, 100 inoculated fungus, sterile distilled water control. incubated at 30 °C 8 days, dried surface wiped to remove exterior signs mold. imaged on both endosperm germ sides over wavenumber range 103–2831 cm−1. mean spectrum extracted from image each kernel, preprocessed adaptive iteratively reweighted penalized least squares, Savitzky-Golay smoothing min-max normalization. Based upon group spectra, 36 51 local peaks identified side embryo area side, respectively. With spectral variables peak locations inputs discriminant models, 3-class principal component analysis-linear analysis models ran 50 random times, achieved overall prediction accuracies 89.47% 75.55% using data, corresponding standard deviations 3.48% 4.34%. results demonstrate usefulness technology differentiating control infected

Correlation and classification of single kernel fluorescence hyperspectral data with aflatoxin concentration in corn kernels inoculated with Aspergillus flavus spores.

The objective of this study was to examine the relationship between fluorescence emissions of corn kernels inoculated with Aspergillus flavus and aflatoxin contamination levels within the kernels. Aflatoxin contamination in corn has been a long-standing problem plaguing the grain industry with potentially devastating consequences to corn growers. In this study, aflatoxin-contaminated corn kerne...

Aflatoxin variation among corn samples with varying ratios of Aspergillus flavus-inoculated/noninoculated kernels.

Aflatoxin levels and physical properties of corn kernels inoculated with Aspergillus flavus during development and noninoculated kernels were compared in samples with various proportions of the 2 kernel types. The relationship between mean toxin levels and associated standard deviations of 5 samples demonstrated a linear association from the lowest toxin in noninoculated corn through a mixture ...

Comparative Histological and Transcriptional Analysis of Maize Kernels Infected with Aspergillus flavus and Fusarium verticillioides

Aspergillus flavus and Fusarium verticillioides infect maize kernels and contaminate them with the mycotoxins aflatoxin, and fumonisin, respectively. Genetic resistance in maize to these fungi and to mycotoxin contamination has been difficult to achieve due to lack of identified resistance genes. The objective of this study was to identify new candidate resistance genes by characterizing their ...

Time course study of substrate utilization by Aspergillus flavus in medium simulating corn (Zea mays) kernels.

Utilization of the three major corn reserve materials, starch, triglycerides (refined corn oil), and zein (storage protein), by Aspergillus flavus was monitored in vitro over a 7-day fermentation. Medium composition in which proportions of reserve materials initially approximated proportions in mature corn kernels changed little over the first 18 h. Subsequently, hydrolysis of both starch and t...

Substrate utilization by Aspergillus flavus in inoculated whole corn kernels and isolated tissues.

Utilization of the major corn (Zea mays) reserve materials (free saccharides, starch, triglycerides, and zein) was monitored during infection of detached kernels by Aspergillus flavus (A. flavus) over a 12-day period. Inoculated whole kernels were compared to noninoculated kernels. Concentrations of sucrose and raffinose in inoculated seed decreased to nearly zero at 6 days, whereas concentrati...
